Have you thought about making your sprites in a vector image editor? That way you can scale them to any size you want before exporting them as pngs or whatever. Granted, they might be a bit cartoony looking, but if you're going for something with casual appeal, that might work best.
Then again, depends on what you're trying to do and what you mean by "casual". :p

Unfiltered things look alright up to about this resolution.
Very artsy looking Doom-resolution sprites (think Hexen/Doom monsters) look absolutely great unfiltered too, but that stuff is very time consuming.

Nah, American football is just silly (e.g. a game of 4x15 minutes that takes around 3 hours to play with more than half the team off the pitch at any one time). It's the other football that rustles my jimmies.
