I always liked opening up to Momento Mori 2's first map. There's always a charm to it that makes me go back to it.

If we're talking all of Doom history, then E1M1. Otherwise, I don't think there's one single PWAD map that stands out the most for me.

Doom's E1M1 is still king in my book. I don't know how Doom 2 managed to screw up it's opening act after the glory that is "climbing stairs to reach armor on a podium," but it did.
Super Mario Bros. 1-1 is also highly discussed among game designers for subliminally teaching the player, but it follows an entirely different game philosophy.
