Having a pen tablet (a good onem like Wacom Intuos) also helps a ton. Although a mouse is usually sufficient.

You'll need something that works with layers. The obvious name is Photoshop, but it's kinda expensive so if you want to stay legit you can try Gimp. http://www.gimp.org/

You could also play around with 2D vector graphics packages like Inkscape and Illustrator. You'll probably still want to do the detailing and final touches in a standard raster package (like gimp, paint.net, photoshop) though.
